As for telling whether a company is sketchy or real (fraud is a nasty word, there’s varying degrees of sketchinees, full out frauds aren’t even worth trading because there’s no shares to short), you gotta dig through the SEC filings (TIMfundamental DVD students know EXACTLY where I look) and try to find inconsistencies in their press releases/conference calls (just like Adam F. did with Hemispherx Biopharma).

A regulatory package that contains data from a phase II study completed 15 years ago in 92 patients and a larger phase III study done five years ago that appears (by my latest count) to have four different sets of results, one of which clearly showed Ampligen failed. (The others indicate the drug barely met its primary endpoint with statistical significance.) Both studies, in fact, used different primary endpoints, and the latter study enrolled patients using outdated criteria for chronic fatigue syndrome.

This is especially odd since Hemispherx claims Ampligen works through a newly discovered mechanism of action not used by any other approved drug. If the FDA is taking Ampligen and chronic fatigue syndrome so seriously, then why hasn't the agency granted fast-track status to the drug? To date, the only confirmed communication made public between Hemispherx and the FDA are two letters sent in 1998 and 2000 by the agency admonishing the company for illegally promoting Ampligen as safe and effective when the drug wasn't approved for sale.

I found that AHRQ report, thanks to the Google. It was published in September 2001 — almost eight years ago. The “most promising” line about Ampligen is in the report, for sure, but it refers to the small Ampligen study done in 1993-1994, not the pivotal study of the drug conducted in 2004.

The report also states that, “In sum, evidence from trials involving immunologic therapies was relatively scant and insufficient to conclude whether these treatments were effective or ineffective.”

Not such a ringing endorsement, but then, Carter doesn’t want investors to know that, nor does he want them to know he’s quoting from an outdated report compiled eight years ago.

Intranasal inoculation with both the annual influenza vaccine and Ampligen(r) may thus represent a strategy to generate protective mucosal immunity against newly emerging and highly pathogenic avian influenza viruses in humans.

Challenging trials clearly demonstrated that vaccine plus Ampligen adjuvant have been most effective versus all other treatment options. It is important to know that Ampligen may be used as an adjuvant for any other future influenza outbreak, not just Swine Flu.

Comments: A new peer reviewed publication titled "Development of mucosal adjuvants for intranasal vaccine for H5N1 influenza viruses" was published in Therapeutics and Clinical Risk Management 2009:5 125-132. The article summarizes ongoing research in which researchers conclude that Ampligen(r)) is the "most promising adjuvant candidate for mucosal vaccines and produced cross-protection against variant influenza viruses not seen with the vaccines given alone.

Is this not the adjuvant task? Indeed it is and Ampligen has done it at best. We need adjuvants and Ampligen is among those whom we really need to have available to infectious disease and oncology specialists.
